Palestine grew with the railroads. Lines arriving in the 1870s stimulated the timber trade, and the Palestine Car Shop is still one of only two car shops on the Union Pacific Railroad, with more than 100 employees. Oil found at Boggy Creek in 1928 diversified the economy, and the city became a center for oil-well servicing and supplies.

Today the largest employer is the Texas Department of Criminal Justice, with more than 3,900 people, and a strong medical and healthcare sector serves a large population of retirees. The 2020 census counted 18,544 residents. U.S. 79, 84 and 287 all pass through town, with Tyler 47 miles to the northeast and Dallas 110 miles to the northwest.
