Port Isabel lies on the western side of the southern Laguna Madre, an estuary of the Gulf of Mexico, 23 miles from the Cameron County seat at Brownsville. Spanish settlers called the place El Frontón; later it became Point Isabel, named for Queen Isabella of Spain. Before the Civil War it was an important port for shipping cotton, and during the war the harbor, the town and its lighthouse changed hands more than once. The city incorporated in 1928 and had 5,028 residents in 2020.

The causeway carries traffic across the lagoon to the island, and the Port Isabel–Cameron County Airport lies northwest of town. The city is counted in the Brownsville–Harlingen–Raymondville metropolitan area and, across the river, the Matamoros–Brownsville area.
